The star is from a run of 1,280,000 notes, yes. While that's not a full run, it's not especially short. I think most people consider a "short run" to be 640,000 or lower (i.e., 20% of a full run or less).

As Noost said, a note that's merely poorly centered is usually considered *less* desirable than a well-centered one. It's only when the centering is so bad that parts of the design are missing, or parts of other notes are showing, that error collectors start to be interested. Even then, where some older notes are concerned, you've got to be aware of the characteristics of each series: For example, the 1928 $1 red-seals are known for very poor vertical centering. I picked this one up at a *discount* because of its bad centering--the tiny sliver of the next note along the top edge is common enough in this series that it's not even called an error:

Even more extreme are the Fractionals (hand-cut at the BEP, as Mpcusa pointed out), and then the Nationals (hand-cut at the individual banks, some of whom were *very* sloppy). I once had a dealer offer me a large-size National for cheap because it had almost half an inch missing at the bottom, with almost half an inch of the next note showing at the top. He said it was "almost enough to be an error"! I've also seen Nationals that were torn, rather than cut, from their sheets....
