This is the 2020 £2 silver proof Mayflower. There is a noticeable smudge at the 8:00 position. Also around the ring where the two metals meet. I have some acetone. Should I follow the typical dipping procedure or use a q-tip w/acetone and try to just get it wet, then wash clean?

I would never use a q-tip on a proof. A bath in acetone can't hurt. After that a quick dip if needed.

I was able to remove the "smudge" with a standard acetone bath and rinse. The residue around the ring didn't change and may be a result of manufacturing.

As predicted. I have posted and you may want to consider what I have said. This is an oxidation that I believe is related to exude from the bonded inner ring surface (this can be simply cleaning agents from the interface and not adhesives per se). But, your coin....

I'll try that for the ring part. Don't take any offense, I had the acetone materials on hand.
