Good eye! Yes thats right. Congrats its missing the mintmark. I'm not sure but I'd suppose it was a grease filled die as you suspect. I'd think the die must of had it, they were putting them on the working hubs by 2000. Its been around the block for sure and its rough but not enough wear to have worn it down or off in my opinion. Its not completely obvious its missing at first glance. when I found it I had to take a triple take on it. I felt something was wrong but couldn't nail down what was the matter at first. Lol.

yes it's unusual in the other denominations it's usually on the front or on the back somewhere on the modern coins except for cents. If it was a proof coin and there were a couple others it would probably be significant, if it was on a BU coin and there were no disruption in the area (suggesting it was left off the die) it would likely be significant. This, I don't really think it's significant besides an oddity of a struck through grease unless a whole bunch have turned up, which hasn't happened to my knowledge in the last 20 years.
I'm just getting my daughter into the state quarters and the 3rd quarter she pulled out if my quarter jar was one of these
