No. There is more than one plate on the press. If you look at a new pack of notes you will see that the back plate numbers rotate.
There are exceptions to this. On occasion one may find pairs of consecutive notes that have identical back plate numbers (and possibly front as well). The typical cause seems to be when a misprinted sheet is removed and destroyed, leaving notes from an adjacent sheet with the same plate numbers to fill their place. It would also seem that the plate numbering process was different in the old days than today. I recently observed three consecutive 1963B $1 Barr notes to have completely identical back and front plate numbers. It remains to be seen if this is an anomaly or if this was the norm for back then.
The earliest 32-subject intaglio presses were single-plate models, producing a stream of sheets that all have the same plate number. I forget when the four-plate presses came along, but it was fairly early, in the '60s or '70s I think. Much more recently, about a decade ago, the BEP acquired several three-plate presses (the SOI presses).
