I just noticed a letter in a fancy font (an A and a B) on these, to the right of the blue 1 and below the IT in United. Does this mean anything? Thanks.

Does plate position have any effect on value?

No, they do not have any effect on value. You will also notice some symbols that can come at the beginning or end of the serial number on large-size notes, but they also do not increase the value of the note.

I agree with Matt but will add that signature combinations can make a big difference. This particular note has 11 different signature combinations, Fr.226 through Fr.236.
