if you have an unopened roll of 2010 coins does that make them have an MS grade?

If it is a US Mint wrapper, then they would all be MS. It's possible that a roll with a wrapper from, say, NF String could be all MS coins, too, but that is just a guess on my part.

I've had a problem with S&S rolls having little black spots on them which would make them not MS. A vast majority are though.

You are going to have to explain that one. Spots might make them corroded or net graded less than MS, but it still does not qualify as wear.

MS just means the coins are uncirculated. If all the coins in the roll are 2010's then they most likely are mint state.
And Lincolncent if your'e talking about cents with the spots, those too are mint state. They do come from the mint with the spots.:(

Per Michael Fahey of ANACS in The Authoritative Reference...: MS60-62 can have "moderate to heavy spotting."

You'll even see GEMs that show a spot or two under magnification.
