On the reverse of the classic Buffalo nickel, directly in front of the Buffalo's front leg/hoof there is a protrusion from the ground.

What is this exactly? A cactus? A pile of rocks?

I can't tell even with magnification and have not come across a reference that explains.

I just never gave it any thought before, but being as large as it is, I wonder what it might be.

Anyone know the answer to this?

Could this be true?! And folks got upset with the 1916 Standing Liberty Quarter? Let's get our ideals in order.

I can't tell you for sure what is in the old Buffalo nickel, but from what I understand the creature was in the New York Zoo where it posed for the coin. So anything might be laying on the ground in New York.
As for what it might be, I can assure you that after herding many of these old Buffs in my life time, it could very well be nothing more than what is called a "Buffalo Pie" Actually quite common all over the pastures where these animals roam. Hope this helps with the puzzle.
